The best way is by XRF or by microscopic cut section imagery.

The most rapid method is the XRF. Cross sectioning takes much more time, but can be used. I'd make my own panels of known nickel in the Zinc alloy. Then, calibrate my XRF machine to assure that the result is accurate and meaningful.
Your XRF supplier may be able to sell you standards. We used to make our own. Plating with Zinc/Nickel, Stripping the coating on Steel, and stripping the coating completely. Then using an A.A., analyze for Zinc and Nickel to derive the exact percentage of each.
